Yugi dedesinin ona verdiği milenyum yapbozunu tamamlar. Bu yapbozu tamamladığı günden beri düello yaparken içinde farklı bir kişiliğin olduğunu hisseder. Bir gün Yugi'nin evine video kaseti ve davetiye gelir arkadaşları dedesiyle izlemeye başlarlar ama bi anda Yugi'nin etrafında ki her şey donar, sadece Pegasus ile kendi kalır Pegasus'la yaptığı düello da zaman yetmediği için kaybeder ve dedesinin ruhunu Pegasus esir alır Yugi'ye ancak düzenlediği düello turnuvasında onu yenerse dedesinin ruhunu serbest bırakacağını söylemesiyle Yugi'nin arkadaşlarıyla macerası başlar.

Yugi Mutou:

Animenin ana karakteridir. Domino Lise'sinde okur. Her zaman okul üniformasını giyer. Yugi'nin saçlarının önlerinde iki yana giden uzun sarı saçları vardır. Saçlarının içi siyah, dışı ise mordur. Büyükbabasının ona verdiği Milenyum Bulmacası'nın parçalarını birleştirdiğinde bedenine 5000 yaşındaki mısır firavunu yerleşir. Firavunun adı Atem'dir. Atem olmadan önce içine kapanık ve yalnız olan Yugi daha sonraları yani Atem geldikten sonra arkadaş edinir. İçine kapanıklığını yener.

Dark Yugi(Firavun Atem,Yami Yugi):

Animenin ana karakteridir. Gizemli güçleri olan eski mısır firavunudur. Milenyum Bulmacası'nın gizemli gücüne sahiptir. 5000 yıl önce Mısır'ı ve Milenyum parçalarını şeytani Zorc'nun elinden kurtarmayı başarır. Tekrar bu olayların olmaması için kendisini ve güçlerini milenyum bulmacasına kitler. 5000 yıl sonra Yugi'nin Milenyum bulmacasının parçalarını birleştirmesiyle olaylar yeniden başlar. Yugi ile hafızasını bulmaya çalışır.

Tea Gardner(Anzu Mazaki):

Yugi'nin en yakın arkadaşlarından biridir ve aynı zamanda sınıf arkadaşıdır. En iyi arkadaşları Yugi Mutou, Joey Wheeler ve Tristan Taylor'dur. Tea genellikle düello yapmayı sevmez ve düello yapma hakkında fazla bilgisi yoktur. Yugi ve Joey azda olsa Tea'ya düello yapmanın nasıl birşey olduğunu gösterir.

Joey Wheeler(Katsuya Jonouchi):

Yugi'nin en yakın arkadaşlarından biridir ve aynı zamanda sınıf arkadaşıdır. En yakın arkadaşları Yugi Mutou,Tea Gardner ve Tristan Taylor'dur. Saç rengi sarışındır ve gözü koyu kahverengidir. Bazen uzun açık yeşil renkli ceketini ve beyaz tişörtünü, kot pantolonunu giyer. Seto Kaiba'nın kendisini aşağılamasından nefret eder ve düellolarda her zaman şanslıdır.

Tristan Taylor(Hiroto Honda):

Yugi'nin en yakın arkadaşlarından biridir ve aynı zamanda sınıf arkadaşıdır. En yakın arkadaşları Yugi Mutou, Tea Gardner ve Joey Wheeler'dır. Düello krallığında önemli bir karakter değildir, fakat savaş şehrinde Joey'in kızkardeşi Serenity'e aşık olur ve onun için Duke ile devamlı yarışır. Serenity için düello yapar.

Ryo Bakura:

Yugi ile aynı sınıftadır. Milenyum parçalarından biri olan milenyum yüzüğüne sahiptir. Beyaz saçlıdır. Milenyum yüzüğü yüzünden kötü tarafı olan Dark Bakura'ya dönüşür.

Dark Bakura:

Aynı Atem gibi 5000 yıllık bir ruhtur ve eskiden firavunun en büyük düşmanlarından biridir ve ruh hırsızıdır.

Seto Kaiba(Kaiba Seto):

Yugi'nin inandığı "Kartların Kalbine" inanmaz. Kahverengi saçlı ve mavi gözlüdür. Yugi ve Joey'i sevmez. Büyü ve gölge düellolarına inanmaz. Büyü ve gölge düellolarını anlamsız bulduğu için devamlı Yugi ve Joey'e "hokus-pokus saçmalıkları" der, fakat Yugi bunların hiçbirine aldırış etmez ve Kaiba'yı arkadaşı olarak görür. Buna rağmen, Yugi'ye yardım eder. Mokuba adında küçük erkek kardeşi vardır ve Mokuba'yı her şeyden korur. Çünkü ailesinden kalan ve onun için değerli olan tek kişidir. Mokuba Kaiba:

Kaiba'nın küçük erkek kardeşidir. Yugi ve arkadaşlarını kendi dostu olarak görür ve onları sever. Annesi öldükten sonra ağabeyi Seto ile öksüzler yurduna bırakılır. Orda Gozaburo Kaiba tarafından evlat edilir. Kaiba Corp'un %2 hissesine sahiptir. Kaiba Corp'ta müdürdür.

Solomon Mutou:

Yugi'nin büyük babasıdır. Gençliğinde bir arkeologtur. Yaşlanınca bir kart dükkanı açar.

Serenity Wheeler:

Joey'in küçük kız kardeşidir. Tristan ile Duke ona aşıktır ve ikisinin arasında kalır. Saf bir kızdır. İlk başlarda kördür fakat Joey, Serenity'i tedavi etmek için Pegasus'un hazırladığı turnuvaya katılır. Serenity daha sonra iyileşir.

Duke Devlin:

Serenity'e aşıktır. Bu nedenle Tristan ile yarışır. Zindan zar canavarları oyununu bulmuştur. Kızlar tarafından sevilen bir kişidir.

Pegasus J. Crawford:

Amerikan kart tasarımcısıdır. İlk başlarda gölge düelloları yapardı. Milenyum gözüne sahipti fakat Yugi'ye düelloda yenilmesinden sonra Bakura tarafından milenyum gözü çalışır. Kaiba Corp'un sahibi olmayı istedi çünkü sevdiği kadın olan Cecilia'yı Solid Vision Teknolojisi'yle geri getireceğini düşündü.

Marik Ishtar:

Mielnyum çubuğunun sahibidir. Ailesi yüzyıllar boyunca firavunun mezar koruyucularıdır. Mezar koruyucu görevine kendisi seçilince içinde kötülük oluştu. Milenyum çubuğunu kullanarak annesini ve babasını gölge diyarına gönderdi. İçinde kötülük büyüdükçe 33 mısır tanrısını bulmak istedi. 3 mısır tanrısını bulmak için Nadide Avcıları adında bir organizasyon kurdu.

Dark Marik:

Marik'in içinde gelişen karanlık kişiliktir. Düello yaptığında karşısındaki rakibini öldürmek için oynar ve bundan büyük bir zevk duyar. Mezar hırsızıdır.

Ishizu Ishtar:

Marik'in kızkardeşidir. Kaiba'yı ve Yugi'yi Savaş Şehri'nde düello yapmaya teşvik eder. Savaş Şehri'nde yarı finale kadar çıkmıştır, fakat Kaiba'ya yenilmiştir. Savaş Şehri'ne katılmasının nedeni Marik'in içindeki kötülüğü yenmektir. Geleceği gösteren Milenyum Kolyesi'ne sahipti, fakat kolyeyi Yugi'ye verdi.

Mai Valentine:

Mai kendinden emin, çekici ve düelloda zorlayıcı bir düellocudur. İlk başlarda sadece kendi için ve yalnız düello yapardı. Sonra Yugi ve arkadaşlarıyla tanışınca bütün duyguları değişir.

Shadi:

Mezar koruyucusudur ve 7 milenyum parçasınında koruyucusudur. Milenyum anahtarının sahibidir.
